Estudou-se a variabilidade genética em couve (Brassica oleracea L. var. acephala D.C.) tipo manteiga por intermédio do polimorfismo enzimático em gel de poliacrilamida e do polimorfismo de DNA, denominado RAPD (Random Amplified Polymorphic DNA), com base na amplificação de segmentos de DNA ao acaso. Avaliaram-se quinze clones de couve-manteiga do Banco Ativo de Germoplasma do Instituto Agronômico (IAC), utilizando-se extratos de folhas para análise de isoenzimas e marcador RAPD com os "primers" dos kits A e B da Operon Technologies. This work was carried out in order to identify Bemisia tabaci (Genn.) biotypes present in the state of Paraná and to determine their geographical distribution and host plants. About 50 adults were collected in several host crops, weeds and ornamental plants in North, Northwest, Northeast, West and Central areas of the state, from January to May, 1998 and 1999. The species were identified by means of RAPD-PCR, using the primer Operon H-16. Whitefly populations were detected mainly from February on, in both years, seldom achieving more than one adult per leaf. The insect was found in only 66% of the sampled areas. Se evaluó la diversidad de especies de Fusarium asociadas con la marchitez del melón (Cucumis melo L.) mediante RAPD, comparándose tales resultados con la identificación taxonómica en base a caracteres morfológicos y patogénicos. Se estudiaron 18 aislamientos colectados de cultivos comerciales de melón establecidos entre la III y IV Región, Chile. Las observaciones morfológicas determinaron que once aislamientos correspondían a F. oxysporum, cuatro a F. semitectum, uno a F. solani y uno a F. subglutinans. Eight Chilean strains of Dunaliella salina obtained within a restricted geographic range, but exhibiting a high variability in their morphology, rate of growth and carotenogenic capacity, were analyzed by Random Amplified Polymorphic DNA (RAPD-PCR). Twenty of the 50 random primers (D, P, OPA and OPD series) that were tested amplified reproducible bands and were useful for comparative analysis of the strains. Of 107 polymorphic genetic markers, 49 were strain-specific. A great genetic variability was found among the strains in spite of their geographic proximity. Five mutants (MaE10, MaE27, MaE24, MaE41 e MaE49) of Metarhizium anisopliae wild strain E9 were analysed for DNA profile through the RAPD technique and for changes in total protein content by spectrophotometry, polyacrylamide gel electrophoresis and densitometry. The pattern of RAPD markers showed genetic polymorphism among the strains: out of twenty primers seven were selected, producing 113 bands. Forty seven bands were present in all strains (41.6% of monomorphic bands) and 66 showed polymorphism (58.4%). The mean coefficient of similarity among all strains was 0.75 (75%). The growth of thirty-four Lentinula edodes strains submitted to different mycelial cultivation conditions (pH and temperature) was evaluated and strain variability was assessed by RAPD molecular markers. The growth at three pH values (5, 6 and 7) and four different temperatures (16, 25, 28 and 37ºC) was measured using the in vitro mycelial development rate and water retention as parameters. Mycelial cultivation was successful at all pH tested, while the ideal temperature for mycelial cultivation ranged between 25 and 28ºC. The water content was lower in strains grown at 37ºC. During some phases of of their life-cycle endophytic fungi colonize plants asymptomatically being found most frequently inside the aerial part of plant tissues. After surface disinfection of apparently healthy leaves from three varieties of mandarin orange and one tangor, and after incubation on appropriate culture medium, 407 fungal isolates were obtained, giving a total infection frequency of 81%. No fungal growth was observed from disinfected seeds, indicating that fungi are probably not transmitted via seeds. The soybean (Glycine max (L.) Merrill) frogeye leaf spot is caused by the fungus Cercospora sojina Hara and is a widespread disease in Brazil and other countries, causing severe losses in grain yield and also affecting seed quality. The availability of DNA markers linked to genes for resistance to this disease would accelerate breeding programs, particularly when other traits are also being evaluated. In the Rio Doce basin of southeastern Brazil, the freshwater fish Hoplias malabaricus (trahira) is a widespread predatory characin and one of the few resilient native fishes in a highly impacted lake system.
